Notes
ICC/IF
1/200.
Dot Blot
Use at an assay dependent dilution. Use at an assay dependent dilution. The
antibody reacts with purified myosin light chains (20,000 daltons) in both the
native protein state or an SDS-heat denatured, reduced state.

Notes
WB
1/5000. Detects a band of approximately 20 kDa. 1/5000 (see review). Detects a
band of approximately 20 kDa. The product shows no reactivity with intact myosin
molecules in their native state. Heat denaturation in the presence of SDS and a
reducing agent (e.g. mercaptoethanol) makes accessible for the monoclonal
antibody an epitope on the myosin light chains.
Target
Function
Myosin regulatory subunit that plays an important role in regulation of both smooth muscle and
nonmuscle cell contractile activity via its phosphorylation. Implicated in cytokinesis, receptor
capping, and cell locomotion.
Tissue specificity
Smooth muscle tissues and in some, but not all, nonmuscle cells.
Sequence similarities
Contains 3 EF-hand domains.
Post-translational
modifications
Phosphorylation increases the actin-activated myosin ATPase activity and thereby regulates the
contractile activity. It is required to generate the driving force in the migration of the cells but not
necessary for localization of myosin-2 at the leading edge.
