Relevance
MAP4K6 is a serine/threonine kinase belonging to the germinal center kinase (GCK) family.
These kinases regulate a wide variety of cellular processes by coupling cell surface receptors to
MAPK pathways. This kinase appears to act upstream of the JUN N-terminal pathway and may
be involved in brain development. It may also play a role in the response to environmental stress,
in cytoskeleton reorganization, cell adhesion, and cell motility. Alternative splicing occurs at this
locus and four transcript variants encoding distinct isoforms have been identified.

Detection of MAP4K6 in Immunoprecipitates
of HeLa whole cell lysate (1 mg for IP, 20% of
IP loaded) using ab86385 at 3 µg/mg lysate
for IP and at 1 µg/ml for subsequent Western
blot detection. Lane 2 represents control IgG
IP. Detection: Chemiluminescence with an
exposure time of 10 seconds.

ICC/IF image of ab86385 stained SKNSH
cells. The cells were 4% PFA fixed (10mins)
and then incubated in 1%BSA / 10% normal
goat serum / 0.3M glycine in 0.1% PBSTween for 1h to permeabilise the cells and
block non-specific protein-protein
interactions. The cells were then incubated
with the antibody (ab86385, 5µg/ml) overnight
at +4°C. The secondary antibody (green) was
goat anti-rabbit DyLight 488 (IgG H+L) used
at a 1/250 dilution for 1h. Alexa Fluor® 594
Immunocytochemistry/ Immunofluorescence -
WGA was used to label plasma membranes
Anti-MAP4K6 antibody (ab86385)
(red) at a 1/200 dilution for 1h. DAPI was
used to stain the cell nuclei (blue) at a
concentration of 1.43µM.
