Description
Mouse monoclonal [6-11B-1] to TUBA4A (acetyl K40)
Specificity
ab11323 recognizes an epitope located on the a3 isoform of Chlamydomonas axonemal atubulin, within four residues of Lys-40 when this amino acid is acetylated. A sequence very
similar to the one detected by ab11323 in Chlamydomonas is found in the majority of a-tubulins,
but the corresponding region is markedly divergent in some a-tubulin isoforms from chicken,
Drosophila and yeast. The antibody has been used to detect acetylated a-tubulins in several
organisms. Occasionally, the epitope recognized by the antibody may be absent or masked as it
is in the kangaroo rat epithelial-like cell line PtK2.

Target
Function
Tubulin is the major constituent of microtubules. It binds two moles of GTP, one at an
exchangeable site on the beta chain and one at a non-exchangeable site on the alpha-chain.
Sequence similarities
Belongs to the tubulin family.
Post-translational
modifications
Some glutamate residues at the C-terminus are polyglutamylated. This modification occurs
exclusively on glutamate residues and results in polyglutamate chains on the gamma-carboxyl
group. Also monoglycylated but not polyglycylated due to the absence of functional TTLL10 in
human. Monoglycylation is mainly limited to tubulin incorporated into axonemes (cilia and
flagella) whereas glutamylation is prevalent in neuronal cells, centrioles, axonemes, and the
mitotic spindle. Both modifications can coexist on the same protein on adjacent residues, and
lowering glycylation levels increases polyglutamylation, and reciprocally. The precise function of
such modifications is still unclear but they regulate the assembly and dynamics of axonemal
microtubules.
Acetylation of alpha-tubulins at Lys-40 stabilizes microtubules and affects affinity and
processivity of microtubule motors. This modification has a role in multiple cellular functions,
ranging from cell motility, cell cycle progression or cell differentiation to intracellular trafficking
and signaling.

fixed and blocked with BSA prior to
incubation with the antibody for 1 hour. An
Alexa-Fluor ® 488 conjugated goat antimouse IgG was used as the secondary.

ab11323 staining Tubulin (acetyl K40) in
human glioblastoma tumor sphere cells by
ICC/IF
(Immunocytochemistry/immunofluorescence).
Cells were fixed with paraformaldehyde,
permeabilized with Triton X-100 0.1% in PBS
and blocked with 0.5% BSA for 20 minutes at
room temperature. Samples were incubated
with primary antibody (1/100 in PBS + 0.5%
BSA) for 1 hour at 4°C. An Alexa Fluor®488conjugated Goat anti-mouse IgG polyclonal
