Product datasheet Anti-S100 antibody - Astrocyte Marker ab868 13 Abreviews 15 References 4 Images Overview Product name Anti-S100 antibody - Astrocyte Marker Description Rabbit polyclonal to S100 - Astrocyte Marker Specificity This antibody recognizes all the S-100 isoforms, proteins of 21-24kDa identified as the A and B subunits of S100 protein. It stains schwannomas, ependymomas, astrogliomas, and almost all benign and malignant melanomas and their metastases. S100 protein is also expressed in the antigen presenting cells such as the Langerhans cells in skin and interdigitating reticulum cells in the paracortex of lymph nodes. Tested applications IHC (PFA fixed), IHC-FoFr, IHC-P, WB, ICC/IF Species reactivity Reacts with: Mouse, Rat, Cow, Human, Pig Immunogen Recombinant full length protein (Cow). Positive control Melanoma or Schwannoma General notes Contains a proprietary preservative that is not sodium azide or thimerosal Properties Form Liquid Storage instructions Shipped at 4°C. PubMed: 20633557 IHC-P 1/100 - 1/200. WB Use at an assay dependent concentration. PubMed: 18032704 ICC/IF Use at an assay dependent concentration. PubMed: 19593677 Target Function Weakly binds calcium but binds zinc very tightly-distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites. Tissue specificity Highly prevalent in heart. Also found in lesser quantities in skeletal muscle and brain. Sequence similarities Belongs to the S-100 family. Contains 2 EF-hand domains. Cellular localization Cytoplasm. Anti-S100 antibody - Astrocyte Marker images ab868 staining mouse brain tissue sections by IHC-P. Sections were formaldehyde fixed and permeabilized in 0.1% Triton-X100 in blocking solution prior to blocking in 100% BioGenex Power Block Universal Blocking Reagent for 10 minutes at 22°C. The primary antibody was diluted 1/1000 in 1% BSA in PBS pH 7.4 and incubated with the section for 1 hour at 22°C. A tyramide Alexa Fluor® 594 conjugate was added and the signal was visualized using a fluorescent microscope and a rhodamine filter 2 ab868 staining S100 in mouse cerebellum by immunohistochemistry (formalin/PFA-fixed paraffin-embedded sections). Cells were paraformaldehyde fixed prior to blocking in 5% serum for 30 minutes at 25°C. The primary antibody was diluted 1/100 and incubated with the sample for 16 hours at 4°C. Alexa fluor® 488 goat polyclonal to rabbit Ig, diluted 1/400, was used as the secondary antibody. Tissue was fixed with paraformaldehyde. Samples were incubated with primary antibody (1/500) for 12 hours at 4°C. An undiluted HRP-conjugated polyclonal was used as the secondary antibody.
