The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user. Application IHC-P Abreviews Notes 1/100. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol. WB 1/5000. Detects a band of approximately 10 kDa (predicted molecular weight: 10 kDa). Target Function Connects the two COX monomers into the physiological dimeric form. Involvement in disease Defects in COX6B1 are a cause of mitochondrial complex IV deficiency (MT-C4D) [MIM:220110]; also known as cytochrome c oxidase deficiency. A disorder of the mitochondrial respiratory chain with heterogeneous clinical manifestations, ranging from isolated myopathy to severe multisystem disease affecting several tissues and organs. Features include hypertrophic cardiomyopathy, hepatomegaly and liver dysfunction, hypotonia, muscle weakness, excercise intolerance, developmental delay, delayed motor development and mental retardation. A subset of patients manifest Leigh syndrome. Sequence similarities Belongs to the cytochrome c oxidase subunit 6B family. Cellular localization Mitochondrion intermembrane space. Anti-Cytochrome C Oxidase subunit VIb antibody [3F9D3D11AF6] (HRP) images 2 Anti-Cytochrome C Oxidase subunit VIb antibody [3F9D3D11AF6] (HRP) (ab198317) at 1/5000 dilution + Human Heart Mitochondrial lysate (ab110337) at 5 µg developed using the ECL technique Performed under reducing conditions. Predicted band size : 10 kDa Observed band size : 10 kDa Western blot - Anti-Cytochrome C Oxidase subunit VIb antibody [3F9D3D11AF6] (HRP) Exposure time : 10 seconds (ab198317) This blot was produced using a 4-12% Bistris gel under the MES buffer system. The gel was run at 200V for 35 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab198317 overnight at 4°C. Antibody binding was visualised using ECL development solution ab133406. 3 IHC image of Cytochrome c oxidase subunit staining in a section of formalin-fixed paraffinembedded normal human heart muscle tissue*, performed on a Leica BOND. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20mins. The section was then incubated with ab198317 at 1/100 dilution, for 15 mins at room temperature. DAB was used as the chromogen. The section was then Immunohistochemistry (Formalin/PFA-fixed counterstained with haematoxylin and paraffin-embedded sections) - Anti-Cytochrome C mounted with DPX. The inset negative control Oxidase subunit VIb antibody [3F9D3D11AF6] image is taken from an identical assay (HRP) (ab198317) without primary antibody. For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times. *Tissue obtained from the Human Research Tissue Bank, supported by the NIHR Cambridge Biomedical Research Centre Please note: All products are "FOR RESEARCH USE ONLY AND ARE NOT INTENDED FOR DIAGNOSTIC OR THERAPEUTIC USE" Our Abpromise to you: Quality guaranteed and expert technical support Replacement or refund for products not performing as stated on the datasheet Valid for 12 months from date of delivery Response to your inquiry within 24 hours We provide support in Chinese, English, French, German, Japanese and Spanish Extensive multi-media technical resources to help you We investigate all quality concerns to ensure our products perform to the highest standards If the product does not perform as described on this datasheet, we will offer a refund or replacement.
