Description
Operation Sequence
The GeneDisc system is a complete, rapid, reliable and easyto-use solution for microbial testing in three simple steps from
sample to result. After samples are extracted with the GeneDisc
Ultra-Lyser, the DNA may be purified by utilizing the GeneDisc
Ultra-Purifier. Extracted and purified DNA is then loaded into a
GeneDisc Plate designed for specific detection and/or quantification of microorganisms of interest. The GeneDisc Plate is then
placed into the GeneDisc Cycler for analysis. The analysis step
by real-time Polymerase Chain Reaction (RT-PCR) runs in as little
as an hour and the final results are displayed with no further
operator intervention.
Purification of DNA Samples
For specific applications, such as quantification of Legionella
in complex waters, DNA purification is a key step of sample
preparation. DNA purification enables:
Step 1: After cell lysis*, transfer lysate to silica column for DNA
binding using multichannel pipette provided.
• To reduce PCR inhibitions due to organic compounds from
samples
• To achieve highly accurate results.
The GeneDisc Ultra-Purifier ensures high quality DNA purification
based on the technology of DNA molecules adsorption to silica
and solvent washings. This protocol, applied to GeneDisc
methods for Legionella, was validated by NF VALIDATION
against NF T90-471 and ISO /TS 12869 standards.
Thus, sample preparation for GeneDisc method provides purified
DNA samples with a high recovery rate, ideal for quantitative
applications.
Step 2: Proceed to DNA washings.
Step 3: Elute DNA molecules.
DNA extracts are now purified and ready to be processed.
Transfer them to the GeneDisc Plate and analyze using the
GeneDisc Cycler.
* Lysis tube rack is supplied with GeneDisc Ultra-Lyser.
