Applications
The AcroPrep Advance filter plate portfolio includes a variety
of membrane types and pore sizes to provide the best
possible performance for a variety of sample preparation
applications. For critical applications, such as multiplexing
and neonatal screening, we have specifically optimized plates
based on the assay fractionation requirements. In addition,
we have a portfolio of plates available for applications such as
general filtration, lysate clarification, nucleic acid binding, and

Performance
Hold-up Volume – Maximizing the amount of available
sample ensures more reliable results and yields more
material available for additional analysis. AcroPrep Advance
filter plates have an improved well design to minimize sample
hold-up, improve sample recovery, and maximize yield.

Our next generation AcroPrep Advance filter plate was
developed with a strong understanding of the most stringent
performance requirements. The result is a plate that provides
improved performance and increased consistency for key
assay parameters. The AcroPrep Advance filter plate
incorporates new membrane sealing technology which
improves sample recovery, filtration rates, and lowers holdup volumes (Figure 1). The newly configured outlet tips
minimize sample weeping during incubations, reduce sample
loss, and minimize hanging drops on the outlet tips following
filtration (Figure 2, 3). Each of these design attributes
improves the performance reproducibility of the plate from
well-to-well, plate-to-plate, and lot-to-lot.

Multiple plates of each type were evaluated for hold-up volume using a
solution of Vitamin B12. After all wells were evacuated, a wash fraction
was collected into a solid bottom plate and read at OD 550 for
concentration of Vitamin B12. AcroPrep Advance: total 9 plates were
tested (3 plates/lot). Competitor M: total 3 plates were tested. All testing
used 96 wells/plate.

Hanging Drops – To help maintain the integrity of samples
and reduce the potential for cross contamination, the outlet
tips of the plate have been redesigned to nearly eliminate the
presence of hanging drops following filtration.

Hanging drops were measured by evacuating wells of fluid, weighing the
plate, and then blotting and re-weighing the plate. Three plates of each
type were tested and the averages calculated.
Weeping – Filter plates should be able to hold sample for
extended periods of times. Weeping, or leaking of fluid from
wells, can occur during incubation periods or after multiple
filter-wash-incubation steps. Pall has engineered the new
AcroPrep Advance filter plates to minimize weeping when
using a variety of fluids including aqueous samples, organic
solvents, and detergent-containing solutions.

Filter plates were filled with 200 µL of the indicated solutions and
incubated over receiver plates for 24 hours. Receiver plates were
checked for the presence of fluid at 1, 2, 3, 8, and 24 hour intervals.
Three plates of each variety were evaluated and the averages calculated.
