Procedure



Turbid broth cultures of Escherichia coli
(24 hr, 37°C in tryptic soy broth) were
diluted 10-5, 10-6, 10-7, 10-8, and 10-9
0.5 ml samples were plated on 3M
Petrifilm (“Total Aerobic” films) and
incubated at 37°C for time intervals
between 6 and 24 hr
Colony counts were determined by
manual and automated methods
Results


Optimal dilution was determined on films
incubated for 24 hr
At 10-7 dilution the colonies remained in the
range of 50 to 80 (0.5 ml volume plated)

The t-test was used to determine if there was
a significant difference between the manual
and automated counts
For 29 films counted:
t = 0.38
There was no significant difference between
manual and automated counts
Conclusions



To detect Escherichia coli colonies on
total aerobic Petrifilm, a minimum
incubation time of 12 hr is required
There was no significant difference
between manual and automated counts
Using automated counting is more rapid
than manual counting, but it does not
decrease the minimum incubation time
needed to form the bacterial colonies
