Production and distribution of diagnostic reagents
Reference carp cell line, virus strain, ethanol-fixed tissue of diseased fish for PCR, plasmid inserted with cDNA
for PCR positive control and ELISA system.
1.
Virus DNA was supplied to Freshwater Aquaculture Center and College of Fisheries Central Luzon State
University, Philippines; Laboratory of virology, Veterinary Research institute of Tunisia; College of Tropical
Agriculture and Human Resources, Department of Molecular Biosciences and Bioengineering, USA; the
Inspection and Quarantine Agriculture, Fisheries and Conservation Department, The Government of Hong
Kong SAR; National Fisheries Research & Development, South Korea; Mediterranean Fisheries Research,
Production and Training Institute, Ministry of Agriculture and Rural Affairs, Republic of Turkey.
2.
Plasmids inserted with cDNA for PCR positive control were supplied to 1 domestic governmental research
institutes.
3.
A cell line of carp fin was supplied to Lab of Aquatic Medicine, College of Veterinary Medicine, Seoul
National University
Part II: Activities specifically related to the mandate
of OIE Reference Laboratories

International harmonisation and standardisation of methods for diagnostic testing or the
production and testing of vaccines
Participated a worldwide interlaboratory ring-test conducted by Dr Niels Jorgen Olesen and Nicole Nicolajsen of
EU Reference Laboratory for Fish Diseases.
4.
Preparation and supply of international reference standards for diagnostic tests or vaccines
1. Reference cell line for virus isolation: KF-1 cell line (originated to Dr R. P. Hedrick, UC Davis, USA), CCB
cell line (originated to Dr M. Neukirch, School of Veterinary Medicine Hanover, Germany), and KKE, CF-1,
NGF-2,-3, LBCB-1,2 and LBCE (originated to our laboratory).

1. Verification of a RT-PCR assay detecting mRNA of the virus
2. Study on fate of the virus in fish and appropriate target tissues for PCR detection of subclinical carrier fish
3. Improvement of ELISA system for detection of carp antibodies against the viral antigens
4. Epizootiological study on the disease among fish population in natural waters
5. Development of more susceptible cell line for the virus isolation and quantification
6. Comparison of pathogenicity to carp among year-on-year isolates.

Transferring cell culture techniques for propagation of KHV on carp cell line and PCR diagnostic techniques to
one researcher of Mediterranean Fisheries Research, Production and Training Institute, Ministry of Agriculture
and Rural Affairs, Republic of Turkey; to one researcher of the Inspection and Quarantine Agriculture, Fisheries
and Conservation Department, The Government of Hong Kong SAR; and to two researcher of National Fisheries
Research & Development, South Korea.
