Introduction to selected aspects of biotechnology
Polymerase chain reaction (PCR): the general principle of the method, steps applications, types of
PCR: RT-PCR; qRT-PCR; multiplex PCR; nested PCR; RFLP-PCR, etc.
Classical methods of nucleic acids’ sequencing: Sanger sequencing, Maxam–Gilbert sequencing,
semi-automated DNA sequencing.
Next-generation sequencing: pyrosequencing, sequencing by synthesis, ligation and reversible
termination.
Detection of unknown mutations (basic research): RFLP; SSCP; ASO, heteroduplex; DGGE, etc.
Detection of known mutations (diagnostics): ASO, PCR-RFLP; DNA microarray, Southern Blot;
Western blot; Northern Blot
Methods of DNA cloning: restriction enzymes; types of vectors; general rules of vectors selection
DNA cloning applications: medicines, agriculture, industry
