Cell Proliferation Assay (MTS)
Promega Cell Titer 96® Aqueous One Solution Cat. No. G3582
200ul aliquots of MTS stock solution are stored at -20C. Thaw on the bench, keep
protected from light!
MTS working solution: 5ul of MTS stock solution per 100ul of culture medium
To determine cell proliferation:
Remove culture medium (wash cells 1x with PBS if the medium was supplemented
with compounds that could alter the pH of the solution)
Per well of a 24-well plate add 200ul of MTS working solution
Incubate cells at 37C for 2h; the colour of the medium will change to purple
Transfer 100ul of supernatant to a 96well ELISA plate and read with a plate reader at
490nm. Background control: empty wells + MTS working solution in triplicate
Cell Viability Assay (MTT)

200ul aliquots of MTT working solution (5mg/ml in PBS) are stored at -20C.
To determine cell viability:
Add 10ul/100ul culture medium of MTT working solution to each well
Incubate cells at 37C for 1-3h
Remove medium and add 200ul of DMSO (per well of a 24-well plate), resuspend
cells and incubate 5 mins at 37C
Transfer 100ul of the solution to a 96well ELISA plate and measure absorbance with
a plate reader at 550nm
