16 nucleotides (must note 3’ & 5’ ends)
o 4 adenosine with phosphate & deoxyribose attached (2 old, 2 new)
o 4 thymine with phosphate & deoxyribose attached (2 old, 2 new)
o 4 guanine with phosphate & deoxyribose attached (2 old, 2 new)
o 4 cytosine with phosphate & deoxyribose attached (2 old, 2 new)
1 DNA helicase (unwinding/unzipping)
1 RNA primase (priming)
1 DNA polymerase (adding bases/proofreading)
1 DNA ligase (joining Okazaki fragments)
1 Nuclease (deletes incorrect nucleotides)
MOVEMENT



You will be divided into two groups of 21. Each group of 21 students must demonstrate a choreographed
representation of the four stages of semiconservative replication using each student as a chemical found during
replication. Your teacher will select 5 group managers which will function as the enzymes in charge of
replication.
o You will model the four stages of replication:
 Step 1: Unzipping/unwinding DNA
 Start with the old twisted strand of DNA joined together with hydrogen bonds between
the bases.
 Helicase will unwind the DNA and break the hydrogen bonds between the bases.
 Step 2: Priming Exposed DNA Strand
 Primase will add a floating/new nucleotide to each of the old strands to act as the
primer.
 Step 3: Adding New Nucleotides
 DNA polymerase will add nucleotides one at a time starting on the 5’ ends. (Create a
mutation for step 4.)
 The leading strand will be formed quickly in one continuous motion.
 An Okazaki fragment will form on the lagging strand because DNA polymerase will fall
off before a complete strand is made.
 Ligase will bring in the missing nucleotide to fill in the gap between the Okazaki
fragments.
 Step 4: Proofreading Completed Strands
 DNA polymerase will “proofread” each nucleotide checking that nucleotides are
correctly matched.
 DNA polymerase will find a mutation signaling nuclease to yank the incorrect nucleotide
from its position.
 DNA polymerase will then add the correct nucleotides and proofread again.
The movement must be succinct, thoughtful, and creative. Each student must know his or her part.
Be as creative as you need to be to enhance your performance (costumes, props, etc.)
REQUIREMENTS




Seamless flow (no abrupt stops)
Music
No talking/gestures
Finished product of two identical strands of DNA (semiconservative replication)
GRADING


Group managers will assign individual intra group grades using a rubric designed by your teacher. (30 points)
Teacher-judges will assign group grades and pick the winning group (extra credit). (40 points)
