Timeline for Generating iPS Cells
Reprogramming Methods and Timeframes
Steps
Isolate fibroblasts from skin biopsies (human
and mouse). Expand and cryo-preserve primary
fibroblasts. Note that other cell types can be
utilized as well. Please contact the Core for
details.
Reprogram primary cells (thawing of cryopreserved cells, transduction with
reprogramming factors, iPS conversion)
Isolate iPS cell colonies and expand cells for
inial characterization
(determine expression of pluripotency markers,
karyotyping mycoplasma testing) or removal of
the reprogramming cassette
AdCre excision to remove reprogramming
transgenes (for lentiviral method only).
l Characterization of iPS cells:
In Vitro differention into Embryoid Body, ICC
for pluripotency markers
Provide iPS cell line to PI (estimated time
frame for the generation of an iPS cell line)
