Applications are invited for a Research Fellow post to be based within the group of Professor Colin
Johnson studying the genetics and molecular cell biology of ciliopathies. Ciliopathies are a class of
human developmental conditions that are of increasing relevance to basic research and clinical
practice. The post is funded by a Medical Research Council project grant and will contribute to the
execution, analysis and interpretation of siRNA-based reverse genetics screens for novel ciliopathy
genes and processes.
The siRNA screens will be performed in collaboration with the BioScreening Technology Group
(BSTG) at the University of Leeds. The project will require an excellent knowledge of modern
techniques of genetics (including next generation sequencing methodologies) and advanced
imaging techniques (including confocal microscopy and high-content imaging). You will also
evaluate screen hits using functional assays, including the assessment of protein-protein
interactions using affinity purification methods (TAP-tagging), so previous experience of these
systems is desirable. A good knowledge of biochemical assays is essential.
Candidates must be enthusiastic, highly motivated and demonstrate a high level of relevant
laboratory experience and technical competence. In addition, the post holder must show the ability
to manage their time effectively and work under pressure, exhibiting effective interpersonal,
communication, and team-working skills. You must have a PhD in molecular/cell biology or a
relevant biomedical subject (or have a date set for examination) and an interest in medical
genetics. A background in human molecular genetics or developmental biology is desirable.

Job Description
Job Summary
The project will require transfection studies with epitope-tagged constructs and siRNA
oligonucleotides, biochemical assays of protein-protein interactions, high content image analysis of
a reverse genetics screen and the anatomical and functional characterization of mouse models of
human ciliopathies. The project will involve cell culture, including derivation of mouse embryonic
fibroblasts, advanced microscopy techniques, recombinant DNA technology, and will have a
significant component of bioinformatic and image analysis.
BioScreening Technology Group (BSTG)
The BioScreening Technology Group (BSTG) provides an interactive service to enable the
characterization of the function and interactions of particular proteins, and support the identification
and validation of potential biomarkers (diagnostic, prognostic or predictive) and therapeutic targets.
More specifically, the Group helps users to develop and formulate plans for the execution of
projects to: (1) develop probes for biomarkers/drug targets and (2) undertake reverse genetic
screens. The BSTG is funded by the Leeds Biomedical and Health Research Centre to promote
collaboration between basic sciences, translational and clinical research. The reverse genetics arm
of the service consist of a manager, postdoctoral fellow and a technician who are all based in the
section of Ophthalmology and Neurosciences.

Main Duties & Responsibilities






Performing data analysis in collaboration with the BioScreening Technology Group (BSTG)
to determine positive ‘hits’ in reverse genetics screens
Analysing next generation sequencing data to identify pathogenic mutations in ciliopathy
patients
Under the guidance of the principal investigator plan and manage your own research
activity (in collaboration with others as appropriate), assisting in designing specific
strategies towards achieving the research goals of the project and managing individual
components of the study
Ensuring good day-to-day progress of the projects and contributing to strategic
development
Designing experimental procedures and technical details
Molecular and cell biology techniques to be used will include:
o DNA/RNA extraction
o PCR for microsatellite genotyping and direct Sanger sequencing
o preparation of libraries for whole exome sequencing
o mammalian cell culture
o transfection with plasmids or siRNA/shRNA
o RT-PCR and quantitative (‘real-time’) RT-PCR
o biochemical assays of protein-protein interactions using affinity purification methods
(TAP-tagging)
o confocal microscopy and live-cell imaging
o high content imaging



















o bioinformatic analyses of data-sets
Generating research output (e.g. publications/presentations) and analysing and interpreting
the results
Communicating and/or presenting research results within the Group and Section
Developing independent and original ideas, as appropriate
Continually evaluating your working practices and protocols and adapting/evolving these
where necessary
Troubleshooting problems with optimisation, assay design, screening, data analysis and
validation and seeking solutions via published literature, company websites and personal

capture and data analyse

Working collaboratively with other staff/students within the Group, BSTG, OPNE and other

national or international conferences as required

preparing proposals and funding applications

GM and COSSH assessments
Contributing to GM, RA and COSSH assessments when appropriate.

Relationships
The post holder will be responsible on a day-to-day basis to the Principal Investigator, Prof. Colin
A. Johnson, through whom s/he will be accountable to the Head of Section (Prof. Chris
Inglehearn), the Director of the Institute and to the Dean of the Faculty of Medicine & Health. S/he
will be expected to interact with other members of the Section of Ophthalmology and
Neurosciences, comprising post-doctoral research fellows, PhD students and research support
staff, and to present data at Section and Institute meetings.

Person Specification
Essential

A PhD in molecular/cell biology or a relevant biomedical subject (or pending with a date
set for examination)

A BSc degree (or equivalent) in human molecular genetics, developmental biology or
related subject

A demonstrable interest in medical genetics and/or developmental biology

Extensive experience in biochemical assays of protein-protein interactions

Extensive experience in cell culture, including culturing primary cells

Extensive experience in siRNA techniques

Practical experience of immunofluoresence microscopy

A proven track record in molecular genetics, including experience of RT-PCR and
quantitative (real time)-PCR, experience in molecular cloning techniques and generation of
vectors

A proven track record of direct sequencing, including whole exome sequencing,
preparative and analysis methodologies

A demonstrative ability to analyse and interpret research results

A demonstrable ability to work unsupervised, with the direction from the Principle
Investigator and as part of the wider research team

Effective communication and interpersonal skills

The Leeds Institute of Cancer and Pathology (LICAP)
Director: Professor Tim Bishop
The Leeds Institute of Cancer and Pathology addresses both laboratory based and clinical
research into cancer with a major focus on translational science. LICAP is one of the largest
cancer Institute’s in the country and has major financial support from the cancer charities. The
laboratories and clinical research are all based on the St James’s site with laboratory activities
being located in the Wellcome Trust Brenner Building and adjacent buildings while the clinical work
is based within Bexley Wing. The Institute consists of seven Sections: Epidemiology &
Biostatistics; Experimental Haematology; Experimental Oncology; Oncology & Clinical Research
(Clinical); Oncology & Clinical Research (Laboratory); Patient Centred Outcome Research; and
Pathology & Tumour Biology.
Leeds Institute of Biomedical & Clinical Sciences (LIBACS) Director: Professor Philip

LIBACS undertakes clinically-driven research from the level of the gene through cellular, tissue and
organ to clinical trials. Our vision is to develop a sustainable centre of excellence for the
advancement of patient care by translating research results into clinical practise and contributing to

Leeds Institute of Rheumatic and Musculoskeletal Medicine (LIRMM), Director: Professor
Paul Emery
LIRMM is dedicated to improving diagnosis, therapy, intervention and outcome across the
spectrum of rheumatic and musculoskeletal medicine. It boasts a dynamic portfolio of research and
education, delivering wide-ranging clinical, translational and basic research across five Sections:
Clinical Musculoskeletal Medicine, Experimental Musculoskeletal Medicine, Clinical Biomechanics

Leeds Institute of Clinical Trials Research (LICTR) Director: Professor Julia Brown
LICTR delivers innovative design, delivery and knowledge transfer in clinical trials research. Our
multidisciplinary approach, in collaboration with basic scientists, clinicians, policy makers,
healthcare providers, public and patients and University colleagues, delivers internationally
competitive research and teaching that makes a significant contribution to the evidence base for
healthcare delivery. The Institutes research is conducted through the Clinical Trials Research Unit
where we have expertise in design and conduct of complex clinical trials incorporating novel
designs to evaluate CTIMPs, complex interventions, diagnostics, medical devices and surgery.
St James’s University Hospital Campus Infrastructure and Facilities (SCIF) Director:

This group covers activities that cover School of Medicine functions for Institutes at St James’s
University Hospital that span more than one institute including biomedical research facilities,
student education, IT, health and safety, estates, seminars, PGR studentships and business
support functions. These functions help support the 5 adjacent buildings on the site.
There are 3 Institutes with staff and students at St James’s: LICAP (Leeds Institute of Cancer
studies and Pathology), LIBACS (Leeds Institute of Biomedical and Clinical Sciences), LIRMM
(Leeds Institute of Rheumatic and Musculoskeletal Medicine). These three institutes are dedicated
to basic, translational, clinical and health research integrated with student education.
