Project: Isolation of minor enterobacteriophages from animal feces and their
characteristics
Work Experience:
Work with bacteriophages: preparing stocks of bacteriophages, purification of phages
DNA from liquid cultures, assaying the DNA content in bacteriophages stocks and
lysates by agarose gel electrophoresis, purification of bacteriophage particles, including
centrifugation through CsCl gradients, purification of bacteriophage particles by
centrifugation through glycerol step gradients, purification of bacteriophage particles by
pelleting/centrifugation.
Molecular cloning: preparation of vector DNA, preparation of DNA to be cloned,
creation of recombinant DNA with DNA ligase, preparing of competent cells,
introduction of recombinant DNA into host organism by chemical transformation and
electroporation, screening for clones with desired DNA by PCR analysis and restriction
analysis, screening for expression of desired genes with SDS-PAGE.
Electron microscopy: preparation grids for electron microscopy, negative staining of
total phage samples with uranium acid, work with transmission electron microscope
Jeol-100X.
