PhD Molecular Genetics and infection
Title: Investigation of the risk factors for development of infection


by continuous ambulatory peritoneal dialysis patients.
Supervisor: Dr. Primrose Freestone, senior clinical lecturer in
microbiology
Project: Renal failure is increasingly being treated by Continuous
Ambulatory Peritoneal Dialysis (CAPD). However, infection leading to
peritonitis and possible CAPD failure is a frequent complication of this
form of kidney dialysis. Infections are often caused by commensal skin
bacteria such as the staphylococci, and Staphylococcousepidermidis and
S. aureusare responsible for over half of all CAPD infections. The host
risk factors that influence CAPD patient susceptibility to infection
currently revolve around issues such as patient hygiene, and little thought
has been given to the role that the host factors released into the CAPD
dialysis fluid may play. We therefore analysed the protein profiles of
CAPD dialysates from a large number of new and established CAPD
patients, as well as examining how the different CAPD dialysates affected
the growth and virulence of bacteria such as the staphylococci. We found
there were significant differences between patient dialysates in terms of
protein profile and how the bacteria grew and expressed virulence factors.
Considered collectively, our data suggests that the host factors released
into the dialysis fluid of CAPD patients could be an additional factor that
influences their chances of developing a potentially life threatening
infection.

SCIENTIFIC SKILLS

Western Blotting, Elisa, SDS-PAGE, PCR and electrophoresis, DNA
preparations, Growth assay, Biofilm, DotSpot, Genetics techniques. FISH,
Karyotyping, DNA sequence.
GENERAL SKILLS

Communication: Good communication skills were gained from giving
presentations, teaching, supervising undergraduates and technicians and
attending seminars and conferences. Also, I have learned to seek and
accept criticism, using this in a positive manner to further build on my
skills. I understand that good communication is vital to a successful
organization and that within training, listening can be more important than
verbal skills.
Team Work: Successful course work was dependent upon participation
and motivation of syndicated project groups. Working towards one goal
with people from other departments and from different backgrounds and
cultures was of my pleasure and added a lot to my experience.
Problem solving and analytical thinking: Having problemsin
establishing and using the right techniques in my PhD research taught me
to think, be creative and find different ways to solve them out.
Computing: Good working knowledge of several word processing
packages and working on getting the international computer driving
licence (ICDL).
Languages: Fluent in Arabic and English.
