Background
Pexidartinib (PLX3397) is an orally active small molecule receptor tyrosine kinase inhibitor that
selectively inhibits the CSF-1 receptor (CSF-1R) as well as the kinase receptors KIT and FLT3ITD. It is used to deplete monocytes which are dependent on CSF-1R signaling. Its chemical
name is: N- [5- [ (5-chloro-1H-pyr rolo[2,3-b]pyr idin-3-yl )methyl] -2-pyridinyl ]-6(trifluoromethyl )-3-pyridinemethanamine, and has the following structural formula:

Hazard Identification
There are limited human data on the overdose of pexidartinib. In 4-week toxicology studies, the
no-observed-adverse-effect levels (NOAELs) of pexidatrtinib were determined to be 10
mg/kg/day in rats and 6 mg/kg/day in dogs. Pexidartinib was shown to cause hepatotoxicity in
clinical trials, including mixed or cholestatic hepatotoxicity, and embryo-fetal toxicity in animal
studies.

· After eye contact: Rinse opened eye for several minutes under running water.

Procedural Materials and Methods:
Prepare 1 g/kg standard mouse chow:
1. Cayman Chemicals CAS Number: 1029044-16-3
a. Mix 0.1 g with 100 g of standard mouse chow.
b. Store chow at 4˚C
Administer to mice
Notify RAR 2 weeks in advance that you plan to supply mice with special chow provided by us..
1. Clearly label the appropriate cages to be used.
2. Label them again once the mice are given PLX3397 chow.
3. Check mice daily and replace chow as necessary.
