Notes
1/5000. Detects a band of approximately 36 kDa (predicted molecular weight: 37
kDa).
IHC-P
1/50 - 1/100. Perform heat mediated antigen retrieval with citrate buffer pH 6
before commencing with IHC staining protocol.
Target
Pathway
Fermentation; pyruvate fermentation to lactate; (S)-lactate from pyruvate: step 1/1.
Involvement in disease
Defects in LDHA are the cause of glycogen storage disease type 11 (GSD11) [MIM:612933]. A
metabolic disorder that results in exertional myoglobinuria, pain, cramps and easy fatigue.
Sequence similarities
Belongs to the LDH/MDH superfamily. LDH family.
Post-translational
modifications
ISGylated.
Cellular localization
Cytoplasm.

IHC image of Lactate
Dehydrogenase staining in a section of
formalin-fixed paraffin-embedded human
hepatocellular carcinoma tissue*, performed
on a Leica BONDTM. The section was pretreated using heat mediated antigen retrieval
with sodium citrate buffer (pH6, epitope
retrieval solution 1) for 20mins. The section
was then incubated with ab202653, 1/50
dilution, for 15 mins at room temperature.
DAB was used as the chromogen. The
Immunohistochemistry (Formalin/PFA-fixed
section was then counterstained with
paraffin-embedded sections) - Anti-Lactate
haematoxylin and mounted with DPX. The
Dehydrogenase antibody [EP1566Y] (HRP)
inset negative control image is taken from an
(ab202653)
identical assay without primary antibody.
For other IHC staining systems (automated
and non-automated) customers should
optimize variable parameters such as antigen
retrieval conditions, primary antibody
concentration and antibody incubation times.

Western blot - Anti-Lactate Dehydrogenase
antibody [EP1566Y] (HRP) (ab202653)
Exposure time : 6 seconds
This blot was produced using a 4-12% Bistris gel under the MOPS buffer system. The
gel was run at 200V for 50 minutes before
being transferred onto a Nitrocellulose
membrane at 30V for 70 minutes. The
membrane was then blocked for an hour
using 2% Bovine Serum Albumin before
being incubated with ab202653 overnight at
4°C. Antibody binding was visualised using
ECL development solution ab133406.
