Target
Function
Potent tight-binding inhibitor of several G1 cyclin/CDK complexes (cyclin E-CDK2, cyclin D2CDK4, and cyclin A-CDK2) and, to lesser extent, of the mitotic cyclin B-CDC2. Negative
regulator of cell proliferation. May play a role in maintenance of the non-proliferative state
throughout life.
Tissue specificity
Expressed in the heart, brain, lung, skeletal muscle, kidney, pancreas and testis. High levels are
seen in the placenta while low levels are seen in the liver.
Involvement in disease
Defects in CDKN1C are a cause of Beckwith-Wiedemann syndrome (BWS) [MIM:130650].
BWS is a genetically heterogeneous disorder characterized by anterior abdominal wall defects
including exomphalos (omphalocele), pre- and postnatal overgrowth, and macroglossia.
Additional less frequent complications include specific developmental defects and a
predisposition to embryonal tumors.
Note=Defects in CDKN1C are involved in tumor formation.

Tissues were from mouse E14.5 wild-type
embryonic heads fixed in neutral buffered
formalin sectioned (5-7 um) horizontally
through the eyes. Antigen retrieval (0.1M
citrate buffer pH6.5) in a 1400W microwave
(5 min high; 5 min low). Sections blocked in
0.4% BSA/5% goat serum in PBS for 1h.
Antibody diluted (1:100) in block soln, applied
overnight at 4C. Detection with anti-Rb Alexa
green Ab (1:500), nuclear counterstain with
Hoechst dye (bisbenzimide).

ab4058 staining mouse E14.5 embryonic eye
tissue sections by IHC-Fr. Sections were
PFA fixed and blocked with 1% BSA for 2
hours at 25°C prior to incubating with ab4058,
diluted 1/100, for 12 hours at 25°C. A FITC
conjugated goat anti-rabbit antibody, diluted
1/400 was used as the secondary.

Heads of mice were removed, fixed in 10%
formalin, dehydrated, embedded in paraffin,
sectioned (5-7 µm).

transgenic embryos were first deparaffinized
and rehydrated. Antigens were retrieved by
microwave treatment in 10 mM Sodium
Citrate buffer (pH 6.0). Following antigen
retrieval, the tissue sections were blocked
with 10% normal horse serum for 30 minutes,
at room temperature. The slides were then
incubated with ab4058 at a 1/250 dilution
overnight at 4°C. Following brief washes in
PBS, the slides were incubated with the
appropriate biotinylated-secondary
antibodies for 30 minutes at 37°C. Antigenantibody complexes were then detected using
streptavidin-linked Alexa 594 (red) at 1/1000
dilution. Sections were mounted using antifade media containing DAPI (blue). Images
were captured using a Nikon Eclipse
